Biography
Dr. Wen got his BSc and MSc in Nankai University. He received his Ph. D in Nankai University in 2002. From 2004 to 2005, he was a post-doctoral researcher in Centre National de la Recherche Scientifique (CNRS UPR9073), Paris, where he identified the novel exonucleolytic maturation of CCA-containing tRNA in Bacillus subtilis firstly. He joined the Institute of Microbiology, CAS as a Principal Investigator in 2005. He focuses on the systems biology and metabolic engineering for amino acids, nucleosides and proteases production in industrial microbes using novel classification and genetic manipulation tools.
Research Interest
Our group develops novel systematic classification and genetic manipulation tools for microbes, with which we focus on the synthesis and regulation mechanism of amino acids, nucleosides and proteases. Systems engineered strains were constructed and the metabolic fluxes in the bacteria were optimized for the maximum production of end-products. Specific interests are: 1.Rapid discrimination of newly isolated Bacillales with industrial applications using Raman spectroscopy and Screening for the stable alkaline protease from isolated wild-type strains and metagenomic libraries. Reveling the mechanism of high activity and stability of alkaline proteases in a structural perspective. 2.Genetic engineering of industrial microbes using novel genetic manipulation toolbox, including transformation methods by combining cell-wall weakening and cell-membrane fluidity disturbing, overcoming the restriction barrier of bacteria harboring multiple restriction-modification systems with the mimicking-of-DNA-methylation-patterns (MoDMP) pipeline, scarless gene knockout and chromosomal site-directed mutagenesis by combining temperature sensitive plasmids and toxic counter-selection markers. 3.Using Corynebacterium glutamicum as a model, study the mechanism of regulation in amino acids biosynthesis pathway and transportation machinery of amino acids. Construct rational designed amino acids-producing strains with systems engineering. 4.With the knowledge of metabolic flux distribution within cells, optimize the downstream fermentation process of amino acids, nucleosides and proteases production, and specifically, developed a two-stage feeding strategy based on the kind and level of feeding nutrients for improving production of L-threonine by Escherichia coli.
