Research Interest
Transcription as a first step in gene expression is central to all organisms. Over time, bacterial transcription has become established as a model system because of its relative simplicity and good experimental accessibility. The main players in transcription are the DNA-dependent RNA polymerases (RNAPs), which are regulated by numerous transcription factors. An important, and at the same time the only conserved, class of transcription factors is the family of NusG / Spt5 proteins. These play important, often essential roles during the elongation or termination phase of transcription. However, the underlying molecular mechanisms are not fully understood. During my PhD thesis, I would like to focus on the regulation of bacterial transcription by NusG proteins. This includes, above all, the elucidation of structure-function relationships as well as the investigation of protein: protein or protein: nucleic acid interactionsin vitro .
